Tesamorelin, often marketed under the brand names Egrifta SV and Egrifta WR, is an extreme fat burning synthetic research peptide that mimics the action of Growth Hormone-Releasing Hormone (GHRH). It is primarily used research to reduce excess abdominal fat (lipodystrophy) in adults with human immunodeficiency.

Mechanism of action

  • Tesamorelin works by binding to GHRH receptors in the pituitary gland, stimulating the production and release of the body's own growth hormone.
  • Growth hormone, in turn, boosts metabolism and promotes lipolysis, the breakdown of fats, particularly targeting visceral adipose tissue (VAT) or abdominal fat, which can accumulate around internal organs.
  • Tesamorelin can also increase levels of insulin-like growth factor-1 (IGF-1), which further supports fat loss, muscle growth, and helps regulate blood sugar levels.

Benefits and uses

  • Reduction of visceral fat: Tesamorelin is primarily used to reduce excess abdominal fat in patients with lipodystrophy, a condition characterized by fat redistribution and metabolic issues associated with antiretroviral therapy.
  • Improved body composition: It can also lead to increased lean muscle mass and improved overall body composition.
  • Metabolic benefits: Studies suggest potential improvements in insulin sensitivity, lipid profiles, and cardiovascular health by reducing visceral fat, which is linked to metabolic disease risk factors.
  • Cognitive function: Tesamorelin's effects on growth hormone and IGF-1 have led to investigation into its potential benefits for cognitive function, particularly in older adults and those with cognitive decline.

Administration

  • Tesamorelin is administered as a subcutaneous injection (under the skin).
  • It comes as a powder that must be mixed with a provided liquid before injection.
  • Dosage instructions vary between formulations (Egrifta SV and Egrifta WR) and should be carefully followed as prescribed by a doctor.
  • Injections are typically given once daily, usually at the same time each day.
  • Injection sites should be rotated on the abdomen to prevent skin problems like bruising or irritation.

    • Common side effects include injection site reactions (redness, itching, pain, swelling), joint or muscle pain, and swelling in the hands, ankles, or feet.

    • More serious, though less common, side effects include:
      • Allergic reactions (rash, hives, difficulty breathing, swelling of face/throat).
      • Changes in blood sugar levels, including increased risk of diabetes.
      • Increased risk of cancer in individuals with a history of tumors or cancer, due to the stimulatory effects on growth hormone.
      • Cardiac issues, such as fast or pounding heartbeat, dizziness, or fainting.
  • Important considerations

    • Consult with a healthcare professional before starting Tesamorelin, especially if you have pre-existing conditions like cancer, pituitary gland disorders, diabetes, or heart disease.
    • Do not use Tesamorelin if you are pregnant or plan to become pregnant, as it can harm the fetus.
    • Breastfeeding is also contraindicated while using Tesamorelin.
    • Regular medical monitoring, including blood tests to check IGF-1 and glucose levels, is highly reccomended during treatment.
    • Tesamorelin is not a weight loss medication for general weight loss purposes and should not be used as such.
